Rapid quantification of total phenolics and ferulic acid in whole wheat using UV–Vis spectrophotometry

Wheat phenolic compounds have benefits of maintaining health and reducing risks some chronic diseases cancers. breeders producers are becoming interested in producing specialty wheat with a higher amount bioactive phenolics, addition to the conventional targets yield, disease pest resistance, milling baking quality, etc. Generally, total content (TPC) is determined using Folin-Ciocalteu reagent, acids quantified through chromatography methods, which relatively time-consuming labor-intensive. In this study, we developed partial least squares (PLS) models for rapid, simplified, accurate determination both TPC ferulic acid whole extract based on spectra data from UV–Vis spectroscopy. The ranged 240 360 nm were used model development. A 60 samples was as calibration set, 20 an external validation set. For model, values R2-calibration R2-cross 0.89. 0.82, 0.78. Compared traditional analytical our approach more convenient rapid achieves comparable accuracy. breeding programs, well other projects focusing that require fast screening selection lines significant compounds, will be greatly benefitted by new method.
